Determining Protein Stability and Structure in One Measurement using Microfluidic Modulation Spectroscopy (MMS).

Abstract:
Protein stability is a critical quality attribute for protein therapeutics and melting temperature (Tm) is most-often used for ranking candidates. Historically, Tm is measured using DSF, DSC, or CD. However, these techniques are one-dimensional and will only give you the Tm. Here we introduce Microfluidic Modulation Spectroscopy thermal ramping to determine not only the melting temperature, but also the protein structure at each temperature, yielding a much more data-rich analysis.
